WARNING: The following contains massive spoilers for the Demon Slayer Manga’s final arc.
Summary
- Developing a multi-layered drug weakened Muzan systematically
- Hybrid drug enabled Corps to draw out final battle with Muzan
- Drugs created by Tamayo and Shinobu rendered Muzan incapable of escaping
Without a shadow of a doubt, Kibutsuji Muzan was undeniably a powerful adversary. Being the first demon who has survived for over a millennium, Muzan boasts immense power and, above all, an extraordinary regenerative capacity, due to his intense will to live.
Despite Muzan’s immense power, he’s not invulnerable to attacks or any drugs given to him. This fact benefits the Demon Slayer Corps, but they must act swiftly as Muzan can expel unwanted substances from his body. Therefore, one may question the importance of the drug that supposedly killed Muzan in ‘Demon Slayer’.
The Development of the Drug That Killed Muzan
Two Unlikely Forces Work Together to Stop the Demon King
As a devoted follower of the Demon Slayer Corps, I’m thrilled to share that the very drugs which ultimately led to Muzan’s downfall were meticulously crafted with the invaluable help of Tamayo, Yushiro, and Shinobu. At first, Tamayo hesitated, but her curiosity and enthusiasm took over when she learned about the opportunity to collaborate with us. You see, she had at last found a remedy that could miraculously transform demons back into humans, making our mission even more achievable.
In collaboration with Tamayo, I helped corner Muzan – the demon I once thought was real and let slip away. Together, we effectively pinned him down.
Although Shinobu harbored intense animosity towards demons, she admired Tamayo and her skills as a physician, which enabled her to craft a far stronger variant of the toxin she’d been developing with the intention of vanquishing Douma. Both characters possess remarkable expertise in their domains: Tamayo understood Muzan’s vulnerabilities and tendencies, while Shinobu was well-versed in Wisteria poison. By combining their unique insights, they devised a complex concoction that would leave Muzan trembling with dread.

Let’s consider developing a combined medication. It would be prudent to assume that this compound may break down under certain conditions.
Over the years, I’ve been part of an intense research journey that led to the creation of a potent pharmaceutical agent. This drug was carefully crafted with the singular goal of slowing down the remarkable regenerative abilities of Muzan, the original demon. Known for his instantaneous healing capabilities, Muzan is an overpowering adversary, bolstered by his extraordinary strength and Blood Demon Art. To aid the Hashira in strategically subduing him, I’ve been involved in formulating a drug that targets his regenerative process.
First Effect
There are several steps involved in the process that made Muzan unable to fight. The drug Muzan had ingested was capable of transforming demons back into humans. Tamayo developed this drug by examining Nezuko’s blood after each battle. Due to the noticeable alterations in Nezuko’s blood, Tamayo managed to formulate a remedy to convert demons into humans.
Tamayo understood that Muzan would break down and eliminate the medication, leading Shinobu to propose concocting a compound drug with various layers. Revealing the primary effect to Muzan turned out crucial, as he spent much of the night examining the impact, eventually overlooking its multi-layered intricacy.
- Tamayo obtained blood samples from multiple high-level demons, which was likely how she invented the Anti-Blood Demon Coagulant.
- The cure for demonization was successfully created during the Swordsmith Village arc.
Second Effect
I’ve always been fascinated by the fact that Muzan has been around for more than a millennium! Yet, during the epic battle, he was at the height of his powers. This is why Shinobu came up with an ingenious idea – to incorporate an aging effect into our potion. With Tamayo’s brilliant concoction, we managed to successfully brew a drug that would make Muzan age 50 years with every passing minute!
Over a span of almost five hours, I watched as the potion coursed through Muzan’s veins, but strangely, it only activated for a meager three hours. This rapid aging process catapulted Muzan into an age over 9,000 years old! The transformation left his once vibrant hair a stark contrast of gray and snow-white. Moreover, it gradually eroded his formidable powers, slowing him down significantly to the point where he could no longer defeat Tanjiro.
Despite his ability to reverse the initial effects, a new challenge emerged—the second effect started consuming his stamina at an alarming rate. This prolonged fatigue left him powerless to counteract it.
The second impact of the drug was probably the most crucial, as it severely weakened Muzan’s power over a three-hour period. This weakness enabled the Hashira to partially subdue Muzan, causing him to transform into a grotesque form and seek refuge within the Infinity Castle until he could dissolve the initial effect. The consequences of this drug exposure even revealed Muzan’s old wounds from the battle fought by Yoriichi, offering the Corps an extra edge in targeting his vulnerable spots.
An hour before sunrise, the effects of the substance within Tamayo started showing on Muzan. Unbeknownst to him earlier, this drug had been causing his aging process to speed up rapidly, resulting in his hair turning gray. It wasn’t until he noticed a slowdown in his movements that he realized something was amiss about his health condition.
Third Effect
During the Sunrise Countdown arc, as daybreak approached, Muzan uncovered more effects of the drug. Since Tamayo had been a trusted ally to Muzan after being transformed into a demon, she was privy to all his vulnerabilities and imperfections, including his fears. Being cowardly, Muzan tried to fragment himself into several parts to evade the Corps. However, an unforeseen consequence of the drug was that it stopped Muzan from splitting himself, thereby reducing his chances of escape as Tamayo had anticipated.
At this phase, the drug was designed with the contingency that if Muzan found himself in a tight spot, he could fragment himself into several parts to avoid dying. It wasn’t until Tamayo observed Muzan dividing himself to dodge death at Yoriichi’s hands, that she became privy to this information.
Fourth Effect
As soon as the third medication kicked in, it was time for the last, crucial drug to target Muzan’s vulnerable spots within his cells. When Muzan tried to flee, his body started to expel blood, signaling the onset of the fourth drug’s influence.
With each medicine taking effect, Muzan noticeably lost strength. The double whammy of aging and deteriorating cells left him exhausted, making it impossible for him to lift his arm. This situation gave an edge to the Hashira, Inosuke, and Zenitsu in their ongoing fight, even though they were also worn out from previous battles. Facing a critical moment, Muzan wrapped himself in a fleshly armor as a last defense, but this only reduced him to an infantile state. In his final move, he tried to possess Tanjiro, hoping for him to carry on Muzan’s will; however, this attempt was futile.
The medicine given to Muzan was part of many tactics used to control him. Realizing they needed a clear advantage to fight Muzan, the Demon Slayer Corps understood this was crucial. Without the drug, Muzan would likely have overpowered them easily. But as the drug gradually weakened and impaired his abilities, the Demon Slayers managed to delay the battle until dawn, which eventually led to Muzan’s death—an outcome he dreaded most.
